THE RESEARCH OF APOPTOSIS OF
SPERMATOGENETIC CELLS IN JUVENILE TESTIS AFTER EXPOSURE TO VINCRISTINE AND
DACTINOMYCIN

Objective: To investigate the
effect of vincristine, dactinomycin and its combination on apoptosis of
spermatogenic cells in juvenile rat and the expression of Fas gene.
Methods: 3-week old male rats were
randomly divided into 5 groups: vincristine-treated group (VG), dactinomyccin
treated group (VDG), smaller dose vincristine and dactinomycin treated
group (sVDG), control group (CG). And all the animals were given
introperitoneal injections of the drugs. HE stain, fluorescent microscope, TUNEL
and immunohistochemical stain were used to detect the apoptotic response of
spermatogenetic cells and the expression of Fas gene.
Results: Significant apoptosis of
spermatogenetic cells were found near or in the lumen of the somniferous tubules
in all treatment groups compared to CG (p<0.01). The incidence of
apoptotic male germ cells of VDG is statistically higher than VG, DG and
sVDG. No increase of Fas protein was observed after treatment with drugs (p>0.05).
Conclusion: Vincristine and
dactinomycin significantly cause apoptotic death of spermatogonia in
juvenile rats. It is position correlation between apoptotic death of
spermatogonia and the category and dosage of vincristine and dactinomycin. Apoptotic
death of spermatogonia may be cause damage of fertility.
