MECHANISM OF L_TYPE CALCIUM ANTAGONIST NIFEDIPINE ON TREATING PRETERM

Objective: To monitor the complex
calcium oscillations in intracellular Ca2+ in first-passage human
myometrial cells after oxytocin stimulation using lasal confocal
microscope. To take a preliminary research on mechanism of L-type calcium
antagonist in treating preterm labour
Methods: After primary cell
culture of human myometrial cell.using desmin and action to assay.we
monitor calcium changes for 90 minutes utilizing lasal confocal microscope
after adding oxytocin and nefidipine.
Results: calcium wave represents
one way to encode cell signal induction Obvious calcium oscillations
occurred after adding oxytocin but calcium waves are irregular and not
synchronizable.nefidipine can oppress calcium oscillations only before the
occurance of calcium wave.
Conclusions: Nifedipine has some
limitation on therpying preterm labour.this implys that there may be other
calcium channel or subunites on myometrial cell membrane.
