THE FUNCTION OF
MDR1 GENE OF PROCTECTION THE BONE UNDERGOING HIGH-DOSE CHEMOTHERAPY

Objective: The study was to improve the effect of
the chemical treatment the Balb/C mice with Hepatocarcinoma. MDR1 gene
transfering into the murine hematopoietic cells of bone marrow and
transplanted into the body of the Balb/C mice to proctection the bone
during the chemical treatment of the Balb/C mice with Hepatocarcinoma.
Methods: Syngeneic Balb/C mice were
subcutaneously injected with murine hepatocellular carcinoma H22 cella for
tumor-bearing mice model. collect the supernatant of PA317-MDR1/A which
packaged a retrovirus vector containing the full length MDR1 cDNA.transfer
the hematopoietic cell of bone marrow by supernatant method.after four days
of cultivation the hematopoietic cell transferred into the MDR1 gene was
imported into the Balb/C mice Hepatocarcinoma.treated them with cyclophosphmide
by the dose of 300mg/kg.to obsive the information of Balb/C mice.
Results: There is significant difference in the
two groups. the 3 Balb/C mice treated with MDR1 gene survived 8 days later
and 3 died. 6 mice all died in the control group.in the group treated with
MDR1 gene survival time of the mice was significant extended (p<0.01).
Conclusion: MDR1 gene transplanted into the body
of the bone the Balb/C mice with Hepatocarcinoma during the chemical
treatment of the Balb/C mice with Hepatocarcinoma can Proctect the bone and
extend the survival time.
