DETECTION OF THE APOPTOSIS AND ITS GENE EXPRESSION IN BONE MARROW CELLS

Objective:
To investigate the apoptosis and its gene
expression in bone marrow cells induced by Human parvovirus B19 (HPVB19).
Methods:
In situ end-labeling (ISEL) was used to detect
the apoptosis in bone marrow cells of 13 patients with aplastic anemia, and
Immunohistochemical method was used to investigate the expression of the
Bcl-2, Bax and Fas gene.
Results: The cell death by apoptosis is observed in 8 HPVB19-DNA
positive patients, the expression of Bax and Fas are present in 3 patients
(3/6). The apoptosis is not found in 5 HPVB19-DNA negative patients, the
expression of Fas is present in only 1 patient (1/5). The expression of
Bcl-2 is not found in 11 patients.
Conclusions:
The HPVB19 may induce the overexpression of Bax
and Fas, thus results in the apoptosis of bone marrow cells.
