A NEW T CELL DEPLETION METHOD USING ATG IN
VITRO

Objective: To prevent graft-versus-host disease (GVHD) following
allogenetic stem cell transplantation, T cell depletion (TCD) strategies
have been used. The effect of TCD with ATG (Anti thymocyte globulin) was
studied.
Methods: 5 ml peripheral blood from each patient with thalassemia (n=10)
was divided into two equal parts. TCD was performed by 5 mg ATG plus
autologous serum as a source of complement into one part; another part for
contrast. CD3+ cells were detected with flow
cytometry for all the blood samples.
Results: CD3+ cells revealed a significantly
reduced from 68% (SD=11%)in contrast group to 5% (SD=1%) in experiment
group (p=0.01).
Conclusion: ATG can effectively deplete T
cell in vitro.
