THE LEVEL OF IL-12, IL-4, IFN-��IN PBMC FROM CHILDREN WITH ASTHMA

Objective: To explore
the relationship PPD stimulated with IL-12, IL-4, IFN-�� level in
children with asthma.
Methods: PBMCs from
24 cases of children with asthma and the 18 cases of health controls were
isolated and were cultured in flat-bottomed 24 well culture plates at a
concentration of 2��106 per well in the presence of PPD (50��g/ml) for 24h
at 370c with 5%CO2 . The level of IL-12, IL-4, IFN-�� in the
culture supernatant was estimated by ELISA.
Results: The amounts
of IL-12 and IFN-��in PBMC with PPD stimulation from asthmatic
patients were greater than those in PBMC with the health controls (P<0.01).
Conclusion: The IL-12
and IFN-��showed lower level in asthmatic patients,
suggesting the Th2 responses is predominant and the problem of
antigen-presenting course existed.
